SAMPLE LOADING AND SET UP FOR BRUKER ASCEND 400 NMR
1) PLACE SAMPLE IN THE DEPTH GAUGE TO MAKE SURE IT IS THE PROPER DEPTH.
2) PLACE THE SAMPLE IN THE CAROUSEL. MAKE NOTE OF WHICH SAMPLE NUMBER YOU INSERT IT IN.
3) MAKE SURE THAT THE LIGHT IS GREEN AND THE HEIGHT OF THE SAMPLE TUBE IS NO HIGHER THAN THE RED SAFETY LEVER ON THE AUTOSAMPLER.
4) FROM THE COMMAND FIELD ON THE TOPSPIN SOFTWARE TYPE “SX #” WHERE THE “#” IS THE NUMBER FOR YOUR SAMPLE AND THERE IS A SPACE BETWEEN SX AND THE NUMBER. IN THIS EXAMPLE IT WAS SAMPLE #1. PRESS ENTER AFTER THE COMMAND.
5) THE AUTOSAMPLER WILL BLINK YELLOW AND LOAD SAMPLE INTO THE MAGNET.
6) THE STATUS OF THE SAMPLE IS INDICATED AT THE TOOLBAR ON THE BOTTOM OF THE SCREEN.
7) ONCE THE SAMPLE IS LOADED GO TO THE “START” TAB ON THE TOP LEFT OF THE SCREEN AND CLICK THE BUTTON FOR “CREATE DATASET”.
8) THE SAMPLE INFORMATION WINDOW WILL POP UP.
9) IN THE “NAME” FIELD TYPE THE NAME OF YOUR SAMPLE. ALSO MAKE SURE THAT THE FIELDS FOR “EXPNO” AND “PROCNO” HAVE A NUMBER IN THEM (USUALLY 1) OR IT WILL GIVE AN ERROR WHEN CREATING YOUR SAMPLE. THESE ARE THE EXPERIMENT NUMBER AND PROCESSED DATA SET NUMBERS FOR YOUR SAMPLE.
10) NEXT GO TO THE EXPERIMENT FIELD. YOU CAN EITHER TYPE IN THE NAME OF YOUR EXPERIMENT IF YOU KNOW IT OR SELECT IT FROM THE LIST BY CLICKING “SELECT”. (IF AN EXPERIMENT IS MISSING OR THE LIST SEEMS ABBREVIATED MAKE SURE THE SOURCE IS C:\BRUKER\TOPSPIN3.2\EXP\STAN\NMR\PAR OR THAT THE BOX LABELLED “SHOW RECOMMENDED” IS NOT CHECKED).
11) NEXT GO TO THE FIELDS FOR SOLVENT. YOU CAN SELECT THE SOLVENT FROM THE DROP BOX. REMEMBER TO CLICK THE BOX FOR “EXECUTE GETPROSOL”.
12) FINALLY FOR THE FIELD “DIR” SELECT THE DIRECTORY TO STORE YOUR DATA. YOU CAN SELECT FROM THE EXISTING FOLDERS OR SIMPLY TYPING IN A NEW NAME AFTER THE “\” WILL CREATE A NEW FOLDER FOR YOU.
13) ONCE COMPLETE CLICK “OK” AT THE BOTTOM OF THE SAMPLE INFORMATION SCREEN.
14) TO EJECT SAMPLE WHEN FINISHED YOUR ANALYSIS ENTER “SX EJ” IN THE COMMANDS FIELD.
